An innovative crosslinking system for controlled, two-step bioconjugation.
Thermo Scientific Bioconjugate Toolkit Reagents provide biomolecule conjugation chemistry that is easy to use and applicable to almost any conjugation scheme. These reagents are heterobifunctional, containing an N-hydroxysuccinimde ester (NHS-ester) and either a hydrazine /hydrazide or an aldehyde group. Biomolecules are first reacted with the NHS ester to form a stable amide bond, followed by subsequent binding to an aldehyde-modified molecule to form a conjugate. This conjugate does not require harsh reducing agents (such as sodium cyanoborohydride) in order to stabilize the conjugate. Conjugates produced utilizing these crosslinkers have extended stabilities in aqueous solution. Figure 1 is a reaction scheme for producing bioconjugates using these new crosslinking reagents.
