n-Dodecyl-b-D-maltoside maltoside is a water soluble nonionic detergent often used in the isolation of membrane proteins. Multiple studies have shown that n-Dodecyl-b-D-maltoside is a gentle detergent that is often able to preserve protein activity better than many commonly used detergents, including Triton x-100, NP-40,CHAPs and Octyl-b-glucoside (1-8).
Like most detergents, n-Dodecyl-b-D-maltoside has dual hydrophobic/hydrophilic properties that allow lipid displacement and provides a lipid-like environment for membrane proteins, however, studies suggest the ability to preserve membrane protein structure by n-Dodecyl-b-D-maltoside, and other gentle detergents, in part stems from reduced disruption of lipid:protein interactions where some of the natural lipid associations are maintained. While no detergent or set of conditions is optimal for all studies on membrane proteins; n-Dodecyl-b-D-maltoside has been shown to be a versatile and reliable detergent.

At concentrations above 0.17 mM n-Dodecyl-b-D-maltoside micelles having MW >50,000. In this form the detergent is difficult to remove by dialysis or gel filtration. Ionic strength of the solution has minimal effect on the CMC.
