Endogen's TriSpot Human IFNγ/IL-2 ELISPOT Kit is a tri-color enzyme-linked immunospot (ELISPOT) assay for the quantitation of single cells releasing both human interferon gamma (IFNg) and interleukin-2 (IL-2). A spot forms at the site where secreted cytokine is bound, is counted (either using a dissection microscope or automated ELISPOT reader) and then compared to the number of cells added to the microplate. This allows the frequency of cells secreting each cytokine to be calculated. One spot is equal to one molecule-secreting cell. IFNγ secretion produces blue spots, IL-2 spots are red and dual secretion is purple.
