SuperSignal West Pico Chemiluminescent Substrate

Thermo Scientific SuperSignal West Pico Chemiluminescent Substrate is an enhanced chemiluminescent (ECL) HRP substrate for low-picogram-level detection by Western blot analysis.

SuperSignal West Pico Substrate provides intense signal and picogram sensitivity for Western blotting with horseradish peroxidase (HRP) enzyme conjugates. The ECL substrate is compatible with different membranes, blocking reagents and a wide range of antibody dilutions. SuperSignal West Pico Substrate provides excellent performance, versatility and economy for routine Western blotting needs.

  • Economical – optimized for dilute antibody concentrations:
    • 0.2 to 1.0µg/mL primary antibody (1:1000 to 1:5000 dilution from 1mg/mL stock)
    • 10 to 50ng/mL secondary antibody (1:20,000 to 1:100,000 dilution from 1mg/mL stock)

Product Details:

SuperSignal West Pico Substrate provides detection of low-picogram levels of proteins in Western blots.
Low-picogram detection with SuperSignal West Pico Substrate. Turbo GFP-His-HA-FLAG was diluted in electrophoresis reducing sample buffer. Lane 1 contained 20pg of Turbo GFP. Seven 1:1 serial dilutions were then prepared and applied at 10μL/well. After electrophoresis, proteins were transferred to nitrocellulose membrane (Part No. 88018) using the Pierce G2 Fast Blotter (Part No. 62288) and Pierce 1-Step Transfer Buffer (Part No. 84731). The membrane was blocked with SuperBlock in TBST Buffer (Part No. 37536). Then the membrane was incubated with Anti-His (Part No. MA1-21315) at 1μg/mL, followed by incubation with Goat anti-Mouse Horseradish Peroxidase Conjugate (Part No. 31430) at 50ng/mL. SuperSignal West Pico Substrate (Part No. 34078) was used for detection. One minute exposures were acquired on CL-XPosure Film, 8 x 10in. (Part No. 34091) and the Thermo Scientific myECL Imager (Part No. 62236). Exposures from the myECL Imager were inverted and contrasted (black=64,000, white=65,535, gamma=1.0).

SuperSignal West Pico Substrate performs well with different lysates, targets, blocking agents, and membranes. Whole cell lysate was serial diluted in reducing sample buffer at 10, 5, 2.5, 1.2, 0.6, and 0.3µg/well with a 15μL/well load and separated by SDS-PAGE. After electrophoresis, proteins were transferred to nitrocellulose membrane (Part No. 88018) using the Pierce G2 Fast Blotter (Part No. 62288) and Pierce 1-Step Transfer Buffer (Part No. 84731). The membrane was blocked with StartingBlock (TBS) T20 (Part No. 37543), 5% non-fat dry milk, or 5% BSA as indicated above. Then the membrane was incubated with Anti-Hsp84 (Part No. PA3-012) at 0.3μg/mL, Anti-HDAC1 (Part No. PA1-860) at 0.5μg/mL, Anti-pan Akt (Part No. MA5-14916) at 1μg/mL, Anti-Erk1 (Part No. MA1-13041) at 1μg/mL, or Anti-Caveolin (Part No. PA1-064) at 0.5μg/mL followed by incubation with Goat anti-Mouse Horseradish Peroxidase Conjugate (Part No. 31430)  or Goat anti-Rabbit Horseradish Peroxidase Conjugate (Part No. 31460) at 20ng/mL. SuperSignal West Pico Substrate (Part No. 34078) was used for detection. Exposures were acquired on CL-XPosure Film, 8 x 10in. (Part No. 34091).

SuperSignal West Pico Substrate versus Luminata Classico Western HRP Substrate
Product comparison. Blots detected using Thermo Scientific SuperSignal West Pico Substrate (top) or Luminata™ Classico Western HRP Substrate (EMD Millipore Corp.) (bottom) at 1 and 5 minutes post substrate incubation. Bands are Hsp86 in HeLa cell lysate samples (10µg total protein in first lane; 2-fold dilutions thereafter), probed with rabbit anti-Hsp86 antibody (Part No. PA3-013) at 1:1000 dilution, followed by goat anti-rabbit HRP antibody (Part No. 31460) at 10ng/mL and exposure to CL-XPosure X-ray film (Part No. 34091).
