Thermo Scientific Pierce D-Luciferin is formulated at greater than 99% purity as both monosodium and monopotassium salts to provide convenience and high performance at a low cost in reporter assays involving firefly luciferase.
Luciferin is the substrate for bioluminescence reactions with firefly luciferase enzyme. Lyophilized potassium and sodium salts of firefly luciferin are stable and easily soluble, and both formulations are compatible with a wide variety of luciferase-based in vivo imaging and in vitro bioluminescent assays. (The decision to use one or the other form is a matter of researcher preference.) Applications include ATP assays, gene reporter assays, high-throughput sequencing and various contamination assays throughout the biotechnology field. For in vivo imaging, the injection of luciferin in model systems enables real-time, non-invasive monitoring of disease progression and/or drug efficacy through bioluminescence.

|Firefly luciferase reaction and example luciferase assay result. Left. Bioluminescent signal from firefly luciferase originates from the oxidation of D-luciferin and requires ATP, oxygen and magnesium. Right. HeLa cells were transfected with Thermo Scientific pCMV-Red Firefly Luc Vector (Part No. 16156) or native firefly luciferase vector, then detected using the Thermo Scientific Pierce Firefly Flash Assay Kit (Part No. 16174) with Signal Enhancer (Part No. 16180).
