Proteomics strategy to investigate S-nitrosylation using mass tags

A new product article describes a method to label, enrich and quantify protein S-nitrosylation using Thermo Scientific iodoTMT™ Labeling Reagents for analysis by mass spectrometry or Western blot.

S-nitrosylation is a post-translational modification (PTM) that regulates numerous processes, including cellular proliferation, apoptosis, smooth muscle relaxation, neurotransmitter release, and differentiation. Ryan Bomgarden and colleagues explain how they used our sulfhydryl-reactive TMT™ reagents to specifically label all cysteine residues or only S-nitrosylated sites in protein lysates and then enriched for tagged peptides to enable multiplex analysis by mass spectrometry or detection of specific proteins by Western blot. The technique is a powerful extension of the classic HPDP biotin-switch assay.
