96-well Filter Spin Plates For Sample Clean-up



Many protein purification and extraction procedures deliver proteins in acidic or basic buffers that typically contain components like imidazole, glutathione, high salt concentrations and detergents. While effective for protein isolation and purification, these buffers are often incompatible with downstream applications. Desalting and detergent removal resins enable researchers to quickly remove these contaminants from protein samples and are available in a 96-well spin plate format for multi-sample and high-throughput sample clean-up procedures.

Zeba Spin Desalting Plates

Removing small molecules, salts and elution agents from a sample can be done by passing a sample through a desalting resin. Desalting resin can also be used to exchange the buffer of a sample. The pre-packed 96-well Thermo Scientific Zeba Spin Desalting Plates do not require resin hydration or dispensing and provide the same high protein recovery as Zeba Spin Desalting Columns, giving at least 95% removal of salts and other small molecules. One Zeba Spin Desalting Plate can process up to 96 samples (20 to 100µL) in 5 minutes.

Protein Recovery and Desalting Efficiency using Thermo Scientific Zeba Spin Desalting Plates. Protein samples (1mg/mL) were prepared in 1M NaCl, and 100µL samples were desalted using Zeba Spin Desalting Plates, 7K MWCO. Protein recovery was determined using the Pierce BCA Protein Assay, and salt removal was determined by conductivity measurements.

Small molecule removal using Thermo Scientific Zeba Spin Desalting Plates. Small samples (0.1mL) of 0.1M DTT or 1mg/mL biotin, FITC or biotin-FITC in PBS were processed through Zeba Spin Desalting Plates, 7K MWCO. Removal of DTT, biotin, FITC and Biotin-FITC was determined using Ellman’s reagent (DTT), HABA/Avidin assay (biotin) and florescence measurements (FITC and Biotin-FITC).

Detergent Removal Spin Plates

Detergents used for protein extraction are difficult to remove from protein samples due to the formation of large micelles. The micelles formed by detergents are large, making the separation of detergents from proteins by size exclusion ineffective. The Thermo Scientific Pierce Detergent Removal Spin Plates are packed with resin that is optimized for eliminating detergent from a sample, which is important for many down-stream applications such as mass spectrometry.

96-well Spin Balance Plate

Efficient detergent removal for high-throughput sample preparation with Thermo Scientific Pierce Detergent Removal Spin Plates. Samples (0.1mL) containing 0.5mg/mL BSA and detergent were processed through the Thermo Scientific Pierce Detergent Removal Spin Plate as described in the protocol. Detergents marked with † were processed using the Pierce Detergent Removal Spin Column, 0.5mL (Part No. 87777) as described in the protocol. The detergent removal and protein recovery properties of each well of the detergent removal spin plate are similar to those of the spin column. Residual SDS was measured using Stains-All; Triton* X-100, Triton X-114 and NP-40 were measured by absorbance at 275nm (protein absorbance was subtracted); sodium deoxycholate, CHAPS, octyl glucoside, octyl thioglucoside and lauryl maltoside were measured using concentrated sulfuric acid and phenol. Removal of Brij*-35 and Tween*-20 was monitored by LC-MS/MS and MALDI-MS analysis. Protein concentration was determined using the Thermo Scientific Pierce BCA Protein Assay (Part No. 23225).
