Zeba Spin Desalting Columns and Devices, 40K MWCO

Thermo Scientific Zeba Spin Desalting Columns and Plates (40K MWCO) contain a size-exclusion chromatography resin that provides excellent protein desalting performance in a centrifuge format for 5μL to 4mL samples.

Zeba Spin Desalting Columns and Plates are polypropylene devices containing a proprietary high-performance size-exclusion resin for effective removal of salts and other small molecules (< 2000 daltons) while providing excellent recovery for protein larger than 40,000 daltons. Available in 5 different spin columns sizes, 96-well filter spin plates, Zeba Desalting Devices provide trouble-free desalting and buffer exchange for sample volumes ranging from 5µL to 4mL. The easy-to-use spin column format dramatically improves results over standard drip-column methodologies and the 96-well spin plate format enables more high-throughput processing.

Zeba Spin Desalting Columns, 40K MWCO contain a proprietary high-performance desalting resin that provides exceptional desalting and protein-recovery characteristics compared to other commercially available resins. Dilute (25µg/mL) protein samples can be successfully processed to achieve greater than 90% retention (removal) of salts and other small molecules (< 2000 MW) while maintaining and good recovery of proteins and other macromolecules (> 40,000 MW). Zeba Spin Desalting Columns can be used to process samples from 5 to 4000µL.

The 40K resin effectively removes salts with excellent protein recovery. Comparison of the 40K MWCO Zeba Desalt Resin with two commercially available resins of similar MWCO (30K-50K). Two concentrations of BSA (66 kDa) were prepared in 1M NaCl. Samples (700µL or 200µL) were applied to 2mL desalting columns pre-equilibrated in water. The columns were centrifuged for 4 minutes at 1,000 x g. Desalted samples were tested for protein recovery (Thermo Scientific Pierce BCA Protein Assay, Part No. 23225) and conductivity. Samples were also resolved by SDS-PAGE and stained with Thermo Scientific Imperial Protein Stain (Part No. 24615) (A) or Pierce Silver Stain Kit (Part No. 24612) (B). The Zeba 40K Desalt Resin resulted in greater NaCl removal and higher protein recovery, especially at low protein concentrations and sample volumes.

The Zeba 96-well Spin Desalting Plates require no resin dispensing or hydration and provide the same high protein recovery as spin columns with at least 95% removal of salts and other small molecules. One plate can process 96 small sample volumes (20 to 100µL) in 5 minutes.
