A protein gel stain for detection with infrared fluorescence imagers.

Krypton Infrared Fluorescent Protein Gel Stain.

Thermo Scientific Krypton Infrared Protein Stain enables sensitive visualization with infrared imagers (680nm excitation; 720nm emission) of proteins separated in polyacrylamide gels or or transferred to membranes.

This fluorescent protein gel stain is for infrared (IR) detection of proteins separated by 1D or 2D polyacrylamide gel electrophoresis (SDS-PAGE). The easy-to-use, high-performance protein stain uses a dye with optimal near-infrared fluorescence that is compatible with the LI-COR* Odyssey* Infrared Imaging System and CCD cameras equipped with infrared filter sets (Ex/Em = 680/720nm). Proteins are detectable at concentrations greater than 0.25ng per band in a typical mini-gel lane (greater than 2ng for the rapid staining protocol). The stain provides high signal intensity, exhibits minimal protein-to-protein variation, has an excellent quantitative dynamic range and is compatible with subsequent destaining and analysis by mass spectrometry.

Product Details:

The primary fluorescent excitation maximum of Krypton Infrared Protein Stain is 690nm. The fluorescence emission maximum is 718nm. For best results, detect bands using infrared laser-based imagers equipped with a 680nm laser light source. A 720nm filter is recommended for emission. Stained gels can be imaged on any platform with the respective excitation and emission filters.

Krypton IR Fluorescent Stain is more sensitive than coomassie dye staining.
Thermo Scientific Krypton Infrared Protein Stain is up to 130 times more sensitive than coomassie dye stains. Proteins were separated in 4-20% Tris-glycine gels and stained with Krypton Infrared Protein Stain or coomassie stain (Thermo Scientific GelCode Blue Stain Reagent, Part No. 24590). The gels were imaged with the LI-COR Odyssey* Infrared Imaging System at 680nm excitation and 720nm emission.


Excellent linearity, uniformity and dynamic range with Kyrpton Infrared Protein Gel Stain.
Thermo Scientific Krypton Infrared Protein Stain produces a linear response to staining with minimal protein-to-protein variation. Relative fluorescent intensity was plotted as a function of protein quantity for proteins of various sizes and containing post-translational modifications. The relative fluorescent intensity for each data point is the average value for triplicate gels.


2D gel staining performance of Krypton Infrared (IR) Fluorescent Protein Gel Stain.
Thermo Scientific Krypton Infrared Protein Stain provides exceptional sensitivity and low background in 2D gel analysis. Processed HeLa cell protein extract (28µg) was focused on a pH 5-8 IPG strip followed by 4-20% SDS-PAGE. The gel was stained using the basic protocol (approx. 2 hours) and imaged with the LI-COR Odyssey* Infrared Imaging System using the 700nm channel.
