Answers to frequently asked questions (FAQs) about Dye Removal Columns


 

Will these columns effectively remove dye from a sample that contains a small protein?
Yes. For best results, use a protein solution at neutral pH (6.5-8.5) with at least 100 mM NaCl to enhance protein recovery. The optimal buffer for purifying proteins < 20 kDa is phosphate-buffered saline (PBS; 100 mM sodium phosphate, 150 mM NaCl; pH 7.2).

Can I use the columns for desalting?
No. The resin is not appropriate for desalting applications.

How many samples can I process with these columns?
The kit includes sufficient material to process 10 samples of 250 µl each for most dyes (see the next question). The kit contains 5 ml of Dye Removal Resin, 10 Spin Columns and 20 Collection Tubes. The ideal labeled-protein concentration range is 1-2.5 mg/ml.

Which fluorescent dyes can I remove with this resin?
DyLight Dyes, rhodamine and fluorescein have been removed successfully using this resin. Samples with dyes in the green spectrum, such as Dylight 549 Dyes and rhodamine, might require double processing because they require a high molar excesses for labeling. When using fluorescent dyes other than those indicated, the ratio of resin to protein/dye mixture will require optimization.

Is there sodium azide in the resin's storage solution?
Yes. It contains 0.02% sodium azide.

Is the resin solvent-compatible?
Yes, the resin is compatible with up to 10% organic solvent such as DMF.

Can the columns be reused?
No. The resin is effective for one-time use only and cannot be regenerated.
