Halt Phosphatase Inhibitor Cocktail safeguards against serine, threonine and protein tyrosine phosphatase activities
Phosphorylation and dephosphorylation is a molecular on/off switch that regulates a number of key biological pathways within the cell, including signal transduction, cell division, and apoptosis. The Halt Phosphatase Inhibitor Cocktail preserves phosphorylation of proteins in cell lysates and tissue extracts. The cocktail contains a mixture of four phosphatase inhibitors of broad specificity, including sodium fluoride, sodium orthovanadate, sodium pyrophosphate and -glycerophosphate. Unlike other commercially available phosphatase inhibitor cocktails which protect against either serine/threonine phosphatases or protein tyrosine phosphatases, the Halt Phosphatase Inhibitor Cocktail protects phosphoproteins from both families of phosphatases (Figure 1). Conveniently packaged in a 100X format, the cocktail is stable at 4°C and is compatible with common protein assays, including the BCA Protein Assay Kit (#23225) and Bradford-based assays such as Coomassie Plus-The Better Bradford Assay (#23236).

Important Product Information
- The Halt Phosphatase Inhibitor Cocktail does not contain protease inhibitors. If necessary, use a broad-spectrum protease inhibitor, such as the Halt Protease Inhibitor Cocktail, in addition to the Phosphatase Inhibitor Cocktail.
- The Halt Phosphatase Inhibitor Cocktail interferes with immobilized metal chelate affinity chromatography and 2-D gel electrophoresis. Either dialyze or desalt sample to effectively remove inhibitors from cell extracts before performing such procedures.
- The Halt Phosphatase Inhibitor Cocktail is supplied as a 100X concentrate that is effective when used at a final concentration of 1X; however, samples that contain particularly high levels of phosphatase activity may require a final concentration of 2-3X.
- The Halt Phosphatase Inhibitor Cocktail is compatible with all Pierce Cell Lysis Solutions.
- The Halt Phosphatase Inhibitor Cocktail is does not affect alkaline phosphatase activity.
